What lies beneath the MDM2-p53 interaction
In DDLPS, excess MDM2 disrupts the guardian of the genome1-4
p53 is a tumor suppressor and functions as the guardian of the genome by responding to oncogenic stress and inducing cell cycle arrest and apoptosis2
p53 is mutated in over 50% of cancers where its tumor suppressor function is compromised4
In tumors where p53 is not mutated, its function can be attenuated by other mechanisms. For example, the function of p53 is disrupted by the presence of excess MDM2 either by its amplification or overexpression5
MDM2 is a negative regulator of p53, and in DDLPS where it is present in excess, MDM2 binds to p53 and targets it for degradation3,4
DDLPS=dedifferentiated liposarcoma.
